use the whole body. For shrimp over 2cm, please take the gill or the pleopods.The virus
quantity is higher in these 2 organs than that in other organs on an infected
shrimp. Taking correct organ for
sampling can encourage higher sensitivity.

What is the grinding method and to what level should I grind the
sample?
|

For grinding
method, mash the samples with Grinder against the tube wall to extrude sample
muscle. This is to break the cell tissues, and release the virus. Normally,
simply place the samples in front of Grinder tip inside the tube and grind the
sample by rotating Grinder for 4~5 times against the tube wall, cell tissue can
be easily broken.
Pressing up
and down the grinder is not recommended.
Not only the efficiency is hardly shown, the reagent liquid might spill
and contamination may occur. The grinding level is to break the visible tissues
and grind it as small as possible.
Turbid, cloudy and even porridge-like liquid will form from the sample
after grinding.

How long can a ground sample be last for? What do I do with the
reagent if the test must be stopped during the process?
|
|
Ground sample
should be used immediately. If the
process must be stopped for some reasons, or Sampling Space is distant from the
Reaction Space, please take the following actions according to the situation.
- Sample can be temporary left in reagent in Tube 0 (Do not transfer
to Tube 1 for diluting) at room temperature. To ensure its accuracy in result,
the test should be continued within 6 hours.
- If Tube 2 and 5 are already dissolved but the reaction must be
stopped, please store the sample in the refrigerator (not in the freezer) and use
it within 2 hours. The rest unused reagents can be left at Sampling Space.
- If Tube 2 and 5 are not yet dissolved, simply store the entire
unused reagent in the refrigerator (not in the freezer) till the resume of
test.
- Before resuming the test, please make sure which step to begin with.
Skip steps which are already done if necessary.
